下调 BIRC2 减轻高糖诱导的人肾小球系膜细胞系HGMCs损伤并促进自噬

Downregulation of BIRC2 attenuates high-glucose-induced injury and promotes autophagy in human glomerular mesangial cell line HGMCs

摘要: 目的 探究含杆状病毒IAP重复蛋白2(BIRC2)对体外人糖尿病肾病(DN)系膜细胞损伤和自噬的影响。方法 常规培养人肾小球系膜细胞系HGMCs,并随机分为对照组、模型组、shNC组、BIRC2 shRNA组和BIRC2 shRNA+YC-1组(HIF-1α抑制剂YC-1,25 μmol/L)。CCK-8法检测HGMCs活性。流式细胞术检测细胞凋亡情况。ELISA法检测各组细胞白细胞介素(IL)-1β、IL-6、肿瘤坏死因子-α(TNF-α)水平。透射电镜观察各组细胞自噬体的数量。Western blot检测各组细胞中BIRC2、HIF-1α/BNIP3/mTOR通路相关蛋白和自噬相关蛋白Beclin-1、微管相关蛋白1轻链3(LC3)的表达情况。结果 与对照组相比,模型组HGMCs的吸光度(A)值(48 h、72 h)、IL-1β、IL-6、TNF-α水平、BIRC2蛋白表达和mTOR磷酸化水平显著上升(P<0.05),HGMCs凋亡率、自噬小体数量、HIF-1α、BNIP3、Beclin-1、LC3Ⅱ/Ⅰ蛋白表达水平显著下降(P<0.05)。与模型组、sh NC组相比,BIRC2 shRNA组HGMCs相应指标变化与上述相反(P<0.05)。结论 下调BIRC2减轻体外人糖尿病肾病系膜细胞系损伤,促进自噬。

Abstract: Objective To investigate the impacts of baculovirus IAP repeat containing protein 2 (BIRC2) on mesangial cell injury and autophagy in human diabetic nephropathy (DN) in vitro. Methods Human glomerular mesangial cells HGMCs were cultured routinely and randomly divided into control group, model group, sh NC group, BIRC2 shRNA group, and BIRC2 shRNA+YC-1 group(HIF-1α inhibitor YC-1, 25 μmol/L). CCK-8 method was applied to detect the activity of HGMCs. Flow cytometry was applied to detect cell apoptosis. ELISA method was applied to detect the levels of interleukin(IL)-1β, IL-6, and tumor necrosis factor-α (TNF-α) of cells in each group. Transmission electron microscopy was applied to observe the number of autophagosomes of cells in each group. Western blot was applied to detect the expression of BIRC2, HIF-1α/BNIP3/mTOR pathway related proteins and autophagy related protein Beclin-1, and microtubule-associated protein 1 light chain 3 (LC3) of cells in each group. Results Compared with the control group, the absorbance (A) value (48 h, 72 h), IL-1β, IL-6, TNF-α levels, BIRC2 protein expression, and mTOR phosphorylation levels of HGMCs in the model group were significantly increased(P<0.05), the apoptosis rate of HGMCs, the number of autophagosomes, and the expression levels of HIF-1α, BNIP3, Beclin-1, and LC3 Ⅱ/Ⅰ proteins were significantly reduced (P<0.05). Compared with the model group and sh NC group, the changes in the corresponding indicators of HGMCs in the BIRC2 shRNA group were opposite (P<0.05). Conclusions Downregulation of BIRC2 expression alleviates the damage of mesangial cells in human diabetic nephropathy and promotes autophagy in vitro.
